Guide to the Services of the Health Science Research Resources Bank
[Cell Lines]

1. How to find cell lines
You can search cell lines registered in Health Science Research Resources Bank (HSRRB) at "Cell Search Format" page in this website. Data base search, animal/tissue-order list, and alphabetical list are available for finding cell lines.
2. How to request cell lines
The Request and Agreement Form for the Supply of Cell Lines (Form A) is to be filled out, signed, and sent to the Health Science Research Resources Bank(HSRRB) by mail or fax. A maximum of five orders can be placed on one sheet. Please use Form A (or its copy) from our website or that attached to the catalog.

6. Precautions in handling cell lines upon receipt

1) Storage
Received cell lines should be stored in liquid nitrogen or cultured immediately after arrival. If the cell lines would be stored in a freezer, they should be kept below -80C. and should not be stored longer than 30 days.
2) Re-culturing procedures
(1) Thawing
Take out the ampule from the styrene foam package and thaw the content within 2 min by shaking in warm (not higher than 37C.) water. Use safety gloves and a face guard to avoid injury from possible explosion of the ampule.
(2) Culturing procedures
All operations should be performed under aseptic conditions. Sterilize the entire surface of the ampule with gauze moist with 70% ethanol or cationic detergent sterilizer, and cut the neck off while wrapped in sterilized gauze. Although every ampule has a groove around the neck for easy cutting off, a precaution may be required to open the ampule because of its hard structure. Transfer the cell suspension to a centrifuge tube, add 10 ml of the medium specified in the catalog, centrifuge the mixture at 1000 rpm for 3 min, and discard the supernatant. Without washing, resuspend the cells in the same medium and culture them by the standard procedure. Unless otherwise specified, the appropriate culture volume would be 5 ml per ampule in a 25 cm2 flask or 6 cm dish for tissue cultures. The cell density given in the attached document or in the catalog may differ from the actual density on thawing. Make sure of cell proliferation before proceeding to passage, because excessive dilution may lead to cell death particularly in hemocytes and lymphocytes.
7. Follow-up surveys
To improve our bank functions, you are kindly requested to fill out the follow-up questionnaire enclosed in the package and return it at your earliest convenience. Please inform us of any unexpected events immediately by fax or e-mail.

Any claims on the quality (especially on the proliferative potential) of the shipped cells should be made within 3 months after shipment.
8. Quality control
The cell lines listed in the catalog are routinely checked on the following three points.
1) Survival rate after thawing
Immediately after thawing, the cells are subjected to the trypan blue exclusion test, and are cultured to confirm proliferative potential.
2) Microbial contamination
Contamination with bacteria, fungi, or mycoplasmas is checked according to the standard operation protocol for quality control prescribed by The Japanese Tissue Culture Association (Tissue Culture Research Communications 9, Supplement 9-11, 1990).
3) Isozyme examination
The species of origin of the cell line is identified by the specific isozyme pattern according to the standard operation protocol for quality control prescribed by The Japanese Tissue Culture Association (Tissue Culture Research Communications 9, Supplement 12-13, 1990).
In addition, DNA finger printing, chromosome tests, and virus checks are carried out in some cell lines.
9. Request for cooperation at the time of publication
Research results obtained from the supplied cell lines can be informative to other researchers. Therefore, we would appreciate receiving a copy of reprint, when you publish the results in journals. We would also like you to state in your paper the JCRB number and name of the cell line used along with the name of this bank as the source.
10. Information services on the listed cell lines
All information on the cell lines distributed by HSRRB is available on the data-base at the National Institute of Biomedical Innovation and open to the public through Internet (http://cellbank.nibio.go.jp/).
You are able to gain access to this data-base by WWW browser. On this data-base you can also find references on the cell lines and photographic data of the quality control experiments (cultured cells, mycoplasma tests, freezing curves, isozyme tests, chromosome tests, etc.) in addition to the items given in the catalog. The methods for the quality control experiments and answers to the questions so far received by the Bank are being accumulated in this data-base.
